Alternatives

Sometimes intracellular staining does not work properly (fluorochrome conjugated monoclonals). The general protocol that was introduced uses fixation and permeabilization of the cells. And some cytokines behave differently than others.
It may be convenient to check living cells and being able to sort these cells on the basis of their cytokine pattern for instance. And maybe you would like to culture these cells after isolation to use in other experiments.
