In general every cell type can be used in the saponin protocol. The only condition to look at the cells in a flow cytometer is a single cell suspension. But cells like erythrocytes will not hold during the procedure. In the saponin step these cells will lyse. Single cell suspensions are a pre but the protocol will also work on slides with sheets of cells, like epidermis. These will be evaluated with use of a microscope instead of the flow cytometer.Since the start of this site the protocol has been used for almost all types of cells.
